Host-microbiota, pro-inflammatory immunity and physiological senescence in wild birds

Triggered by microbial ligands, inflammation serves as a "double-edged sword" to fight infections on the one hand, but on the other hand causing tissue damage due to oxidative stress if it is dysregulated. For example, chronic inflammation can contribute to inflammaging, which is now widely regarded as one of the causes of ageing. In my interdisciplinary dissertation, my colleagues and I investigated three interrelated aspects of inflammation, using an evolutionary framework and various free-living birds as models: (1) ecological and evolutionary determinants of gut microbiota (GM) composition and diversity, a driver of wild bird immunity, (2) diversity in immune genes affecting inflammatory responses in wild birds and (3) inflammation-related physiological senescence in a free-living passerine bird, the great tit (Parus major). Firstly, using 16S rRNA gene metabarcoding, we revealed high intra- and interspecific variation in passerine gut microbiota (GM) dominated by the major phyla Proteobacteria, Firmicutes, Actinobacteria and Bacteroidetes. Although in mammals GM depends strongly on host phylogeny and diet, in birds we found only moderate effects of phylogeny and very limited effects of host geography and ecology on GM composition. Utilisation of fluorescence techniques for analysis of industrially relevant microorganisms

Techniky analýzy jednotlivých buněk neboli single-cell analýzy, jsou velmi důležité v kontextu studia důležitých biotechnologických mikroorganismů. V biotechnologických procesech je optimální mít rychlé, efektivní a real-time analýzy, které lze provést s minimální přípravou vzorku. Proto jsme v rámci předložené disertační práce využili autofluorescenci jako marker fyziologického stavu mikrobiální kultury, který je možné analyzovat pomocí fluorescenční mikroskopie a/nebo průtokové cytometrie. Nejdříve bylo potvrzeno, na základně emisních spekter, že zelená autofluorescence emitovaná bakteriemi Cupriavidus necator H16 a jejím polyhydroxyalkanoáty (PHA) neprodukujícím mutantem Cupriavidus necator PHB-4, pochází skutečně z flavinů. Emisní maximum bylo naměřeno v rozmezí 510–550 nm pro FAD, FMN a riboflavin – bohužel jednotlivá spektra od sebe nebylo možné rozeznat, a 515 nm pro oba bakteriální kmeny Cupriavidus necator. Byly nalezeny dvě maxima excitačních spekter, jedno v rozmezí 360–370 nm a druhé okolo 440 nm. Z těchto poznatků byl sestaven nový protokol pro fluorescenční mikroskopii – laser byl nastaven na hodnotu 467 nm a emisní filtr pro detekci byl vybrán 520/35 nm. Flaviny a jejich autofluorescence může být take použita v kombinaci s jinými fluorofory, když jsou použity multiparametrické analýzy, ale je důrazně doporučeno, aby vybrané fluorescenční sondy buď emitovaly v jiném rozmezí, než je 500–550 nm anebo byla jejich doba života excitovaného stavu jiná, než je interval 3,2–3,7 a 4,2 – 4,9 ns. Tyto doby života navíc zůstaly konstantní, i když byly buňky vystaveny kultivačním a stresovým podmínkám. To z nich dělá velmi stabilní marker v porovnání s dynamickým charakterem intenzit fluorescence. Dále byl optimalizován protokol pro značení bakterie C. necator s využitím PHA specifické sondy BODIPY 493/503. Optimální koncentrace této lipofilní sondy byla stanovena na 2,5 µg ml-1 a sondu je možné dle našich dat aplikovat společně s viabilitní sondou propidium iodidem. Tento protokol byl také využit pro studium morfologie PHA-syntetizující halofilní bakterie Halomonas halophila. Pro stanovení UV-protektivních vlastností PHA obsaženého v bakterii Cupriavidus necator, byl optimalizován další protokol, tentokrát pro ROS citlivou sondu CM-H2DCFDA, která byla stanovená jako citlivější v porovnání s její nemethylovanou formou. V této studii byly navíc potvrzeny UV-protektivní vlastnosti PHA na bakterii Cupriavidus necator H16 v porovnání s PHA nesyntetizujícím kmenem Cupriavidus necator PHB-4.

Acute and chronic morphine administration can significantly reduce ischemia- reperfusion injury of the rat heart. However, the molecular mechanisms mediating the protective effect of morphine are not yet fully elucidated. Concurrently, there is a lack of information about the effects of the long-term action of morphine on heart tissue. Therefore, in the first part of the project, we studied the effect of long-term administration of high doses of morphine (10 mg/kg/day, 10 days) on rat heart tissue. In the second part of the project, we investigated the effect of 1 mM morphine on viability and redox state of rat cardiomyoblast cell line H9c2 that was influenced by oxidative stress elicited by exposure to 300 μM tert-butyl hydroperoxide (t-BHP). Our experiments have shown that long-term morphine administration affected neither the amount nor the affinity of myocardial β-adrenergic receptors (β-AR), but almost doubled the number of the dominant isoforms of myocardial adenylyl cyclase (AC) V/VI and led to supersensitization of AC. At the same time, proteomic analyses revealed that long-term morphine administration was associated with significant changes in the left ventricular proteome. Změna abundance mRNA, oxidativních produktů a aktivit antioxidačních enzymů spojených se stárnutím rybích oocytů

In fish, delayed spawning in nature, delayed egg collection in capture and delayed fertilization after egg stripping lead to excessive oocyte ageing and finally over-ripening. Until now there has been only poor understanding about the processes and underlying mechanism of oocyte ageing in fish as well as other vertebrates. Some studies on other vertebrates have suggested the oxidative stress as initiating factor for the progress of oocyte ageing. The present work investigated possible changes in the mRNA levels of transcripts involved in oxidative damage, mitochondrial function and stress response, with roles in fertilization, embryo development, transcriptional regulation and cell cycling as well as the ones related to the apoptosis during the egg over-ripening. This study investigated the oxidation status of oocytes during post-ovulatory ageing by measuring TBARs as the marker of lipid oxidation and carbonyls which shows the extension of protein oxidation. In addition the role of oxidative stress in the progress of oocyte ageing was assessed by evaluation the activity of antioxidant enzymes, CAT, SOD, GPX and GR. Possible changes in lipid class and fatty acid composition during fish oocyte aging were also examined. Egg viability parameters and larval ploidy levels were examined. Complete loss of egg viability for African catfish (Clarias gariepinus) occurred at 16 and 24 Hours Post Stripping (HPS) when eggs were stored at 25°C and 4°C respectively. Under both storage temperatures, the embryo mortality and larval malformation rates increased significantly over time and were the highest in the most aged oocytes. We determined the time in which the eggs retain their fertilizing ability 10 hours for tench (Tinca tinca) at 20 °C, 18 hours for goldfish (Carassius auratus) at 20 °C and 14 hours Post Ovulation (HPO) and more than 10 Hours Post Stripping (HPS) for common carp (Cyprinus carpio) at 20 °C. Our results demonstrated no significant changes in the mRNA levels of oxidative stress related genes and genes involved in cell cycling during the progress of oocyte ageing in any of species; African catfish, goldfish and common carp. Additionally, with elapsing time following ovulation the amount of TBARs and carbonyls, did not change in any of the oocytes from our experimented species; African catfish, tench, goldfish and common carp. However an increase in the mRNA abundance of apoptotic related genes were observed. Antioxidant enzyme analysis indicated no significant changes in the activity of CAT and SOD during post-ovulatory ageing of common carp, goldfish and tench oocytes. However, a significant decrease in the activity of GPX was observed during post-ovulatory ageing of tench oocytes which could play a major role in the overall drop of egg quality. This observation in GPX activity was not observed in common carp and goldfish oocytes. Besides the direct effects of oxidative stress on oocytes, also changes of fatty acid and lipid composition caused by oxidation could affect the functionality. We observed that post-ovulatory ageing of the oocytes does not change the fatty acids and lipid class composition of the oocytes. Therefore, according to our obtained results, oxidative stress is not the main initiator or promotor of the oocyte ageing process. However, complementary tests and analysis are required to clearly clarify its involvement. Increased mRNA levels of apoptotic related transcripts during oocyte ageing in this study demonstrates that apoptotic pathway might be involved in the molecular changes during the progress of oocyte ageing. Investigation of epigenetic changes associated with fish oocyte ageing seems to be interesting for future research works.
The impact of opioids on the effect of cytostatic agents on the C6 and CCF-STTG1 astrocytoma cell lines

Despite its numerous side effects, morphine and the opioids derived from this drug, belong among the only effective options for treatment of pain linked to oncological illness. The effect of opioids on the efficiency of cytostatics in vitro has been the subject of many papers but the results are often contradictory, which could be probably caused by the great variability of experimental models and approaches. Some recent studies indicate that the consequences of activation of opioid signaling in astrocytes display certain differences from other cell types. Glioblastoma multiforme, the tumor derived from astrocytes, belong among those with the worst prognosis, mostly for the frequent resistance to cytostatics. In this thesis we focused on the effect of morphine, methadone and DADLE on the efficiency of cytostatics of temozolomide, doxorubicin and vincristine on the cell lines C6 and CCF-STTG1 derived from glioblastomas. Also, we examined the effect of the above mentioned opioids on the level of oxidative cellular stress and using N-acetylcysteine, a ROS scavenger, we verified the role of oxidative stress in cellular systems activated by the effect of the mentioned opioids on the efficiency of cytostatics. Dita Kašparová Chronická hypoxie a exprese genů 4 Abstract Adaptation to chronic hypoxia (CH) is characterized by a variety of functional changes in order to maintain metabolic and energy homeostasis. It has been known for many years that both humans and animals indigenous or adapted to high-altitude hypoxia are more tolerant to an acute ischemic injury of the heart. Cardioprotective mechanisms activated by adaptive responses to chronic hypoxia can be the result of altered transcriptional regulations in left ventricles. Here we report results from the gene expression profiling of adaptive responses in three models of chronically hypoxic heart. Adult male Wistar rats were exposed for 21 days to either continuous normobaric hypoxia (CCH; 10% O2) or CCH interrupted daily by 1-hour reoxygenation (RCH) or CCH interrupted daily by 16-hour (CIH). Cardiprotective effect of CCH adaptation is abolished by brief daily reoxygenation, RCH adaptation.
